To pa predstavlja tudi osnovo za morebitno smer zdravljenja raka, so danes sporo\u010dili s Kemijskega in\u0161tituta.<\/p>\n<p>Celice reagirajo na spremembe v okolju preko prenosa signalov, ki spro\u017eijo procese, kot je npr. vnetni odziv. Pri tem je pomemben protein MyD88, saj igra klju\u010dno vlogo v signalizaciji imunskega odziva, do katerega pride pri oku\u017ebah in drugih po\u0161kodbah. Po drugi strani pa pretirana aktivacija celic lahko privede do bolezni.<\/p>\n<p>Prva avtorica raziskave Mateja Man\u010dek Keber \u017ee ve\u010d let raziskuje vlogo zunajceli\u010dnih veziklov. Ti so neke vrste mehur\u010dki, zamejeni z lipidno membrano, ki vsebujejo sestavine celic, kot so proteini, oligosaharidi, lipidi in RNK.<\/p>\n<p>Zunajceli\u010dne vezikle je najti v krvi in drugih telesnih teko\u010dinah in njihov pomen je zlasti v vlogi posredovanja informacije med izvornimi in tar\u010dnimi celicami, zlasti pa se je to pokazalo pomembno pri raku.<\/p>\n<p>V Waldenstr\u00f6movi makroglobulinemiji, vrsti limfoma, raka krvnih celic, so odkrili, da ve\u010d kot 90 odstotkov pacientov vsebuje mutiran protein MyD88 L265P, ki povzro\u010di od receptorjev neodvisno aktivacijo celic.<\/p>\n<p>Blokiranje signalizacije preko tega proteina povzro\u010di smrt rakavih celic in o molekulskem mehanizmu aktivacije so raziskovalci Kemijskega in\u0161tituta poro\u010dali \u017ee leta 2014.<\/p>\n<p>Raziskovalci so pokazali, da se MyD88 L265P, prenesen z zunajceli\u010dnimi vezikli, sprosti v citoplazmo tar\u010dnih celic, kjer tvori signalni kompleks z lastnim MyD88 in spro\u017ei vnetje. Vnos zunajceli\u010dnih veziklov so opazili v mi\u0161ih, kjer so zunajceli\u010dni vezikli spremenili mikro okolje v kostnem mozgu.<\/p>\n<p>MyD88 so zaznali tudi v zunajceli\u010dnih veziklih, ki so bili izolirani iz kostnega mozga bolnikov z Waldenstr\u00f6movo makroglobulinemijo, kar po navedbah in\u0161tituta dokazuje, da do tega procesa prihaja tudi v pacientih z rakom.<\/p>\n<p>Avtorji raziskave so Mateja Man\u010dek Keber, Du\u0161ko Lain\u0161\u010dek, Mojca Ben\u010dina s Kemijskega in\u0161tituta, Rok Romih z In\u0161tituta za biologijo celic Medicinske fakultete Univerze v Ljubljani, Jiaji G. Chen, Zachary R. Hunter in Steven P.
